WebJun 9, 2011 · A darkfield microscope is a compound microscope which uses an aperture in the shape of an annulus that is placed between the light source and condenser lens. Ring-shaped light that passes through the aperture is focused by the condenser onto the biological specimen or material sample to be observed. WebThis principle is applied in darkfield (also called darkground) microscopy, a simple and popular method for making unstained transparent specimens clearly visible. Such objects often have refractive indices very close in value to that of their surroundings and are difficult to image in conventional brightfield microscopy.
